Painful diabetic polyneuropathy, defined as pain caused by lesions of the somatosensory system, is attributed to diabetes. Gabapentin is commonly used to treat diabetic neuropathic pain, and targets the a2d unit of the voltage gate calcium ion channel (VGCC). Gabapentin has also been shown to attenuate the inflammatory component that is related to diabetes. This study suggests that the activity of gabapentin is regimen-dependent, and demonstrates a MoA of the cross-talk between the inflammatory process and gabapentin effect in the spinal cord.
